Molecular Sequence of Melanotan-2 Peptide
Ac–Nle–c[Asp–His–D-Phe–Arg–Trp–Lys]–NH₂
- Ac = Acetyl group at the N-terminus
- Nle = Norleucine (a methionine analog)
- c[ … ] = The sequence is cyclized between Asp and Lys
- D-Phe = D-Phenylalanine (non-natural isomer)
- NH₂ = Amidation at C-terminus
Handling and Storage of Melanotan-2 Peptide
The peptide compound should be stored in a cool and dry place to maximize its stability. For Melanotan-2 lyophilized peptides, it is recommended to store them in the refrigerator between 2°C and 8°C (36°F–46°F) in an airtight container.
Lab user exposure:
- Avoid prolonged or repeated exposure.
- Avoid inhalation.
- Avoid contact with eyes or skin.
- Handle inside standard laboratory containment.
- Use gloves, a lab coat, and protective eyewear.
Redissolving:
- Can be reconstituted in sterile distilled water or suitable buffer solutions.
- A small amount of DMSO or 0.1% TFA may be used—use only one at a time to assess solubility.
- Mix gently, avoid high-temperature heating.
Choice of container and precautions:
- Low-binding sterile polypropylene to prevent absorption
- Borosilicate glass vials to prevent absorption in the glass walls
- Avoid polystyrene
- Avoid lab plastic containers
Overview of the Melanotan-2 Peptide
Melanotan-2 (MT-II) is a synthetic cyclic heptapeptide designed as an analog of the naturally occurring peptide α-Melanocyte Stimulating Hormone (α-MSH). In preclinical research, Melanotan-2 is being studied for its potential effects on melanocortin receptor signaling pathways in laboratory models.
Working Mechanism of Melanotan-2 Peptide
Preclinical studies suggest that the compound may help in activating melanocortin receptors:
- MC1R,
- MC3R,
- MC4R,
- MC5R,
- Laboratory studies suggest that (MT-II) may act through the MC1R receptor, which is present on skin melanocytes, to increase intracellular cAMP levels and activate protein kinase A in experimental models.
- According to laboratory experimental studies, Melanotan-2 may enhance the expression of microphthalmia-associated transcription factors. This factor promotes the activities of enzymes such as tyrosinase involved in melanin production in laboratory models.
- Preclinical studies suggest that MT-2, may also augment the synthesis of eumelanin-the dark, protective melanin form-resulting in increased eumelanin production in research models.
TFA Salt Form
Melanotan-2 is provided in its TFA salt form. Residual trifluoroacetate content is typically up to 10% w/w. It depends on the final formulation and lyophilization conditions. The exact TFA content can be confirmed by batch-specific COA.
