Molecular Sequence of Triptorelin (GnRH Peptide)
Triptorelin (Gonadotropin-Releasing Hormone agonist)
Amino Acid Sequence (10-mer Peptide)
pGlu–His–Trp–Ser–Tyr–D-Trp–Leu–Arg–Pro–Gly–NH₂
pGlu-His-Trp-Ser-Tyr-D-Trp-Leu-Arg-Pro-Gly-NH₂
- pGlu (Pyroglutamic acid) at the N-terminus
- D-Trp substitution at position 6 (this increases potency and stability)
- C-terminal amidation (-NH₂) for longer half-life
Handling and Storage of Triptorelin (GnRH Peptide)
The peptide compound should be stored in a cool and dry place to maximize its stability. For Triptorelin (GnRH Peptide) lyophilized peptides, it is recommended to store them in the refrigerator between 2°C and 8°C (36°F–46°F) in an airtight container.
Lab user exposure:
- Avoid prolonged or repeated exposure.
- Avoid inhalation.
- Avoid contact with eyes or skin.
- Handle inside standard laboratory containment.
- Use gloves, a lab coat, and protective eyewear.
Redissolving:
- Can be reconstituted in sterile distilled water or suitable buffer solutions.
- A small amount of DMSO or 0.1% TFA may be used to test or improve solubility.
- Mix gently, avoid high-temperature heating.
Choice of container and precautions:
- Low-binding sterile polypropylene to prevent adsorption
- Borosilicate glass vials to prevent adsorption in the glass walls
- Avoid polystyrene
- Avoid lab plastic containers
Overview of the Triptorelin (GnRH Peptide)
According to preclinical studies, Triptorelin is a synthetic decapeptide (10-amino-acid peptide) that mimics the natural hormone GnRH, which is produced by the hypothalamus in experimental models. In analytical models, GnRH regulates the release of LH (Luteinizing Hormone) and FSH (Follicle-Stimulating Hormone) from the pituitary gland.
When Triptorelin is first administered, it binds strongly to GnRH receptors in the anterior pituitary.
Working Mechanism of the Triptorelin (GnRH Peptide)
This causes a sudden increase (“flare”) in the release of:
- LH (Luteinizing Hormone)
- FSH (Follicle-Stimulating Hormone) in experimental models.
According to preclinical studies, this stimulation lasts only a short time (usually 5–10 days).
- Desensitize the GnRH receptors
- Internalize or “turn off” these receptors
- Stop responding to GnRH altogether
This stage is called down-regulation. After about 2–3 weeks, the pituitary gland stops releasing LH and FSH.
Triptorelin first temporarily increases LH/FSH, but continuous exposure shuts down GnRH receptors in the pituitary, leading to long-term suppression of testosterone, estrogen, and progesterone.
TFA Salt Form
Triptorelin is provided in their TFA salt form. Residual trifluoroacetate content is typically up to 10% w/w. It depends on the final formulation and lyophilization conditions. The exact TFA content can be confirmed by batch-specific COA.
